Why supercritical CO2 could soon replace environmentally damaging solvents in key experimental procedures

27th January 2023

Organic solvents that are commonly used in various chemical processes have significant financial costs and can be environmentally damaging. Therefore, a key goal for both industrial and academic labs is to reduce their use in favour of green solvent alternatives. Here Paul Vanden Branden, director and product manager at laboratory equipment supplier SciMed, discusses the potential of supercritical CO2 for replacing environmentally damaging solvents, such as hexane, in separations and extractions.

Using supercritical liquid CO2 in experimental procedures is becoming increasingly appealing because it is a green solvent. It is non-flammable, non-toxic, not environmentally damaging, easy to prepare and does not contribute to global warming as any process CO2 can be recycled in the system.

Environmentally benign CO2 holds significant potential in its supercritical fluid phase to reduce the reliance on expensive, hazardous and environmentally harmful organic solvents such as hexane. While suitable for use in a range of applications, this is particularly appealing in procedures such as chemical extractions, separations and purifications. Supercritical fluid extraction (SFE) is an area of growing interest and CO2 is the most common supercritical fluid used in SFE procedures.

The benefits of SFE procedures

SFE is performed by pumping supercritical CO2 through a fixed bed of substrate where the CO2 flows through the substrate and dissolves soluble components until they are depleted. The loaded solvent is then passed through a separator where the soluble components are precipitated by adjusting the temperature and pressure before the CO2 is condensed and recirculated.

By controlling the density of supercritical CO2 fluids, they can replicate the performance of various organic solvents whose polarities range from n-pentane, at the lowest density, to pyridine, at the highest. This feature allows selective extraction, purification and fractionation procedures to be performed.

A particular area of interest is using supercritical CO2 as a replacement for hexane, which is typically used for solvent extractions even though it is known to be environmentally harmful. Hexane is widely used for extracting oil contaminants from water and soil for analysis and for extracting edible oils from seeds and vegetables because of its efficiency in these experiments, which could not traditionally be matched by alternatives. However, supercritical CO2 systems now allow comparable results to be achieved without using such a harmful chemical.

Globally, regulations are tightening around the use of harmful solvents such as hexane, with both the UK’s Department of Trade and Industry and the USA’s Environmental Protection Agency publishing recommended procedures using supercritical CO2. Therefore, it makes sense for industry and academia to switch to supercritical CO2 systems and procedures now, before regulations ultimately ban harmful solvents altogether, which is the likely final outcome.

SciMed exclusively represents Core Separations, the world’s leading supercritical CO2 company, which produces systems for laboratory-scale applications (around 10 ml) and production-scale applications (250 to 500 litres). Furthermore, its team of experts can work with you on proof-of-concept testing for any new SFE procedures that you want to develop.

This is just one of the many ways that laboratories can improve sustainability while improving experimental output and saving money. Other methods include using equipment that reduces the use of utilities and increasing the use of field-based analytical instruments to reduce the reliance on outsourcing to commercial laboratories.

Scientific study validates field-based method of measuring oil contamination

12th December 2022

Field-based non-dispersive infra-red spectroscopy instrument produces results that were previously only possible using lab-based techniques.

A new scientific report by Concawe, which reviewed field-based and lab-based methods of analysing oil-contaminated soils, determined that the InfraCal 2 ATR-SP total petroleum hydrocarbon (TPH) analyser is an effective instrument for measuring oil contamination of soil and water. The review showed that the InfraCal 2 performs well for the detection and quantification of TPH between 100 and 10,000 mg kg-1, independent of soil type and fuel type. This allows operators to conduct in-depth sample analysis in the field, meaning they can accurately identify hydrocarbon levels without relying on outsourcing to commercial laboratories.

Historically, measuring oil contamination in soils was done using a combination of non-specific field-based screening tests and specific lab-based fingerprint studies, carried out off-site by a commercial laboratory partner using certified techniques.

While the lab-based techniques, such as gas chromatography coupled to mass spectrometry, provide in-depth data into hydrocarbon speciation and offer high accuracy and precision, they are generally expensive and time-consuming compared to on-site analysis.

However, despite the drawbacks of lab-based techniques, they have been necessary, until now, because field-based techniques could not provide specific data on hydrocarbon speciation and, instead, only provided insight into hydrocarbon concentration hotspots.

Paul Vanden Branden

Concawe’s report confirmed the InfraCal 2, which is a non-dispersive infra-red spectroscopy instrument, is a versatile, fast and easy-to-use instrument for conducting on-site analysis of oil contaminants. For example, it demonstrated how the InfraCal 2 can accurately perform TPH determination of hydrocarbons in five to ten minutes, significantly faster than lab-based GC-MS.

“Developing field-based instruments that compete with lab-based techniques for analysing oil-contaminated soils has been a major industry focus for some time,” explained Paul Vanden Branden, director and product manager at SciMed, the UK’s largest distributor of scientific instrumentation. “For an instrument to be highlighted in a scientific review for performing analysis to a standard that was previously only possible using lab-based techniques reassures businesses that modern field-based instruments are up to the job.

“Using the InfraCal 2 to conduct TPH analysis on site without outsourcing further analysis to commercial labs will save businesses time and money without sacrificing results. Furthermore, it will help companies lower their carbon footprints as it will cut down on sample transportation requirements and minimise the use of harmful chemicals,” concluded Vanden Branden.

The School of Geography, Geology and the Environment (SGGE), University of Leicester have recently taken delivery of a Rigaku Primus IV WD-XRF spectrometer and supported by SciMed ltd.

Key to the success was the advanced features of the instrument for mapping down to Boron and the ease of operation of the software.

Flexibility of the analytical methods to combine calibrations with standardless analysis allows researchers to work with difficult materials and produce results with the minimum of setup time.

This new instrument joins a suite of analytical equipment (incl. advanced microscopy, SEM, LA-ICP-MS and others) in an effort to modernise and advance research support capabilities.

The Primus IV will facilitate rapid, non-destructive, multi-element chemical analysis/characterisation of solid samples including rocks, soils, ceramics, metals, and alloys and is typically used as the first level of data generation for a multitude of research applications. Specifically, within SGGE, this includes investigations into the fundamental Earth processes that contribute to the development of economic ore-deposits, in addition to the development of novel, environmentally friendly critical-metal extraction methods.

The Primus will also contribute to research more broadly across the College of Science and Engineering. The introduction of its new light element analytical capabilities and small spot mapping will prove essential in the assessment of innovative coatings technologies, including the assessment of thickness and evenness of distribution on a variety of alloy surfaces.

It is envisaged that this new system with offer enhanced synergy across SGGE analytical capabilities, facilitating a ‘one-stop-shop’ approach in the macro to micro chemical characterisation of research materials.

The instrument is now installed and SciMed are working to bring the instrument into full use. SciMed look forward to a process of continuous development with The University of Leicester to enable the University to obtain maximum benefit for the instrument with support and knowledge from our expert team.

Industry magazine “Pharma Manufacturing” awarded the Multi N/C 2100 S Pharma analyser from Analytik Jena with the 2020 Pharma Innovation Award in the “Monitoring and Analytical Devices” category

Vaccine Quality Control

In pharmaceutical vaccine production the starting, intermediate and finished products all need to be controlled for the levels of antigens. The total protein analysis in the production of vaccines by determination of the total nitrogen content according to e.g. USP 1057> Method 7.2, enables the quantification of these antigens.

The Analytik Jena Multi N/C 2100 S Pharma TN analyser is the only analyser specifically designed for protein determination in vaccine quality control; designed with catalytic high-temperature combustion and chemiluminescence (CLD) detection for efficient protein analysis in vaccine production, or TN cleaning validation, with minimal sample consumption and a high degree of automation.

  • No sample carry-over due to its unique septum-free direct-injection.
  • Minimum sample consumption with a typical injection volume of between 50 – 200 μl.
  • Savings on costs for expensive products e.g. in vaccine quality control.
  • Large working range from 0 – 200 ppm Total Nitrogen.
  • High degree of automation for large sample sequences (up to 112 sample positions for 2ml HPLC vials).
  • Unattended 24/7 operation thanks to self-check system.
  • File Protection for full data integrity and 21 CFR part 11 compliance.
